چکیده

The physicochemical characterization of the textile industry effluent collected from Oswal Textile Industries, Ludhiana (Punjab.) India has been carried out and the results showed that the temperature (40°C), pH (8.00), Biological Oxygen Demand (260 mgl –1 ), Chemical Oxygen Demand (790 mgl –1 ), Total Suspended Solids (2000 mgl –1 ), Total Dissolved Solids (7000 mgl –1 and colour over the prescribed fresh water limits. A potential bacterial strain was isolated and selected from the textile effluent on the basis of rapid azo dye Crystal violet (100mgl –1 ) decolorization and later identified as belonging to genus Bacillus based on Phenotypic characterization and phylogenetic analysis of the 16s rRNA gene sequence. Effects of physicochemical parameters (pH, Temperature, etc.) on the Crystal violet decolorization by the Bacillus were studied. Decolorization was effective at pH 8, 35°C with starch and peptone as carbon and nitrogen sources and in static conditions. This decolorization potential increased the applicability of this microorganism for the dye removal.

Biological Control of African Violets Root-Knot Disease by the Used of Extracellular Protease Bacillus

The present study explored the efficacy of Bacillus spp. and protease production for biocontrol of the root-knot nematode Meloidogyne javanica in African violet media. Among 100 bacterial isolates from various soils, the highest nematode mortality was observed for treatments with isolate GM-18, which was identified as Bacillus subtilis based on cultural and morphological characteristics and 16S...

Heterologous expression and characterization of laccase 2 from Coprinopsis cinerea capable of decolourizing different recalcitrant dyes

The gene (CcLcc2) encoding laccase from the basidiomycete Coprinopsis cinerea Okayama-7 #130 was synthesized by polymerase chain reaction-based two-step DNA synthesis, and heterologously expressed in Pichia pastoris. The recombinant protein was purified by ammonium sulphate precipitation and nickel nitrilotriacetic acid chromatography. The molecular mass of CcLcc2 was estimated to be 54 kDa by ...

Cellular responses of Bacillus subtilis and Escherichia coli to the Gram stain.

Exponentially growing cells of Bacillus subtilis and Escherichia coli were Gram stained with potassium trichloro(eta 2-ethylene)platinum(II) (TPt) in place of the usual KI-I2 mordant. This electron-dense probe allowed the staining mechanism to be followed and compared with cellular perturbations throughout the staining process. A crystal violet (CV)-TPt chemical complex was formed within the ce...

Experimental Studies on Decolourization of Azo Dye in Textile Effluent by using Bio Remediation Technique

Studies were carried out on the decolorization of azo dyes by Bacillus species from effluent of textile industries. About 10 bacterial strains were isolated from the effluent of textile industries, Bacillus sp. showed remarkable ability in decolorizing the widely utilized azo dyes. Phenotypic characterization and phylogenetic analysis based on 16S rDNA sequence comparisons indicate that these s...
